What Semax is
- A heptapeptide — the ACTH (4–7) fragment with an attached Pro-Gly-Pro sequence that stabilises the molecule.
- Deliberately built so that it **has none of the hormonal activity of ACTH**, that is, it does not stimulate the adrenal cortex.
- Developed in Russia, where it is authorised as a medicine for some indications.
Documented mechanism
- Published work describes an effect on the expression of neurotrophic factors, particularly BDNF and NGF.
- Studied in connection with a modulatory action on dopaminergic and serotonergic signalling.
- In models, a neuroprotective effect in ischaemic injury is described.
- The Pro-Gly-Pro triad at the end is, according to the literature, the reason for its relatively good stability compared with other short peptides.

State of knowledge
Semax has an extensive literature, but predominantly Russian and not always
available in international peer-reviewed databases. Independent replications by
Western institutions are rare. Authorisation in one jurisdiction does not mean
efficacy demonstrated to the standards required by the EMA or the FDA.
The efficacy and safety of administering this substance to humans are not sufficiently established clinically. The information given here is solely a summary of published research findings, predominantly from laboratory and animal models. - Run the solvent down the wall of the vial, not directly onto the lyophilised powder. Do not shake the vial — swirl gently until fully dissolved.
