Description
- A heptapeptide — the tuftsin fragment with an attached Pro-Gly-Pro sequence that stabilises the molecule. The same construction as Semax.
- Tuftsin itself is a natural four-residue peptide derived from the IgG heavy chain, described in connection with macrophage function.
- Developed in Russia, where it is authorised as a medicine for some indications.
- Supplied as a lyophilised powder for reconstitution.
- Published work describes an effect on GABAergic signalling — but, unlike the benzodiazepines, not through the benzodiazepine binding site, so the sedation and dependence characteristic of that class are not described.
- An effect on the expression of neurotrophic factors, particularly BDNF, is reported.
- A separate line of work concerns slowed breakdown of enkephalins: Selank is described in the literature as inhibiting the enzymes that cleave leu-enkephalin.
- In models an effect on cytokine expression, particularly IL-6, is also described, which authors link to the molecule’s origin in immunologically active tuftsin.
- The Pro-Gly-Pro triad at the end is, according to the literature, the reason for its relatively good stability compared with other short peptides.
- The anxiety and stress response in animal models.
- GABAergic signalling without the benzodiazepine binding site.
- Neurotrophic factors and neuroplasticity.
- Enkephalin metabolism.
Selank has an extensive literature, but predominantly Russian and not always available in international peer-reviewed databases. Independent replications by Western institutions are rare. Authorisation in one jurisdiction is tied to the requirements there and does not correspond to the EMA or FDA route.
